So unless your phone is overly large odds are a connected speaker will be able to produce a better range of sound then the small speaker in your phone. That being said audio quality is determined generally by the lowest quality component in the chain. So using a $30,000 pair of headphones to listen to a mp3 ripped out of a youtube video would be pointless. my guess would be unless your phone is exceptionally crappy your built in audio processing is probably good enough to be better then the best the NDS game can produce and a decent speaker/pair of headphones should make sure you are getting the best possible audio experience you can get.
